Identification of Vicia spp. in Republic of Korea by DNA Barcode Gene

Abstract

Vicia spp. was one of the weed occurred in orchard although they can be used as cover crop. To identify simply and clearly by using barcode sequence, rbcL partial gene and internal transcribed spacer (ITS) region of 5 Vicia spp. were sequenced. There was no intraspecific variation in the rbcL partial sequence except V. amoena as 0.1% where the intraspecific variation of the ITS region of V. amoena and V. angustifolia were 0.6 and 0.4%, respectively. Interspecific variation among the five Vicia spp. was 1.1-1.9% in the rbcL and 3.3-3.9% in the ITS region. Based on neighbor-joining tree using the rbcL and the ITS sequences of the 5 Vicia spp., they were clearly grouped suggesting that the rbcL partial gene and the ITS region sequences could be useful to identify Vicia spp. occurred in upland and orchard. Clear identification of weeds in the crop field could help to manage appropriately along with physiological characteristics of weeds.
