Establishment of In vivo Bioassay Using Giant Duckweed (Spirodela polyrhiza L.) Turion for an Efficient Screening of New Herbicidal Compounds

Abstract

This study was conducted to develop In vivo high-throughput screening (HTS) system using giant duckweed (Spirodela polyrhiza L [SPIPO]) turion for new herbicidal agents. In order to increase the reliability of the screening method, it was appropriate to select and inoculate the best turion, and to use dimethyl sulfoxide of 0.5% or less for dissolution of the compound. Based on the results of this study and previous research, basic screening formats using 48 and 96 well plates were established after settling of preparation of turion and chemical solutions, optimal culture conditions, and evaluating criteria of herbicidal activity and symptoms. Then, 26 representative herbicidal agents with different mechanisms of action were tested for a verification of this system. Finally, In vivo HTS for new herbicidal agents named “SPIPO turion bioassay” and its standard protocol was established. This screening method had several advantages such as a use of turions with genetic uniformity, rough distinction of an action type of screening substances, diversification of the experimental conditions, an acquisition and management of the data for various purposes, relatively simple methods and low costs of experiment tools, etc. and seemed to be very useful for a rapid screening of herbicidal substance with new action site, especially.
